Deconstruction of cellulose, the most abundant plant cell wall polysaccharide, requires the cooperative activity of a large repertoire of microbial enzymes. Modular cellulases contain non-catalytic type A Carbohydrate-Binding Modules (CBMs) that specifically bind to the crystalline regions of cellulose, thus promoting enzyme efficacy through proximity and targeting effects. Although type A CBMs play a critical role in cellulose recycling, their mechanism of action remains poorly understood. Here we produced a library of recombinant CBMs representative of the known diversity of type A modules. The binding properties of 40 CBMs, in fusion with an N-terminal green fluorescence protein (GFP) domain, revealed that type A CBMs possess the ability to recognize different crystalline forms of cellulose and chitin over a wide range of temperatures, pHs and ionic strengths. A Spirochaeta thermophila CBM64, in particular, displayed plasticity in its capacity to bind both crystalline and soluble carbohydrates under a wide range of extreme conditions. The structure of S. thermophila StCBM64C revealed an untwisted, flat, carbohydrate-binding interface comprising the side chains of four tryptophan residues in a coplanar linear arrangement. Significantly, two highly conserved asparagine side chains, each one located between two tryptophan residues, are critical to insoluble and soluble glucan recognition but not to bind xyloglucan. Thus, CBM64 compact structure and its extended and versatile ligand interacting platform illustrates how type A CBMs target their appended plant cell wall degrading enzymes to a diversity of recalcitrant carbohydrates under a wide range of environmental conditions.

This paper focuses on the analysis of InGaZnO thin-film transistors (TFTs) and circuits under the influence of different temperatures and bias stress, shedding light into their robustness when used in real-world applications. For temperature-dependent measurements, a temperature range of 15 to 85 • C was considered. In case of bias stress, both gate and drain bias were applied for 60 min. Though isolated transistors show a variation of drain current as high as 56% and 172% during bias voltage and temperature stress, the employed circuits were able to counteract it. Inverters and two-TFT current mirrors following simple circuit topologies showed a gain variation below 8%, while the improved robustness of a cascode current mirror design is proven by showing a gain variation less than 5%. The demonstration that the proper selection of TFT materials and circuit topologies results in robust operation of oxide electronics under different stress conditions and over a reasonable range of temperatures proves that the technology is suitable for applications such as smart food packaging and wearables.

We show that a physical trigger, a non-ionizing infrared (IR) radiation at wavelengths strongly absorbed by liquid water, can be used to induce and kinetically control protein (periodic) self-assembly in solution. This phenomenon is explained by considering the effect of IR light on the structuring of protein interfacial water. Our results indicate that the IR radiation can promote enhanced mutual correlations of water molecules in the protein hydration shell. We report on the radiation-induced increase in both the strength and cooperativeness of H-bonds. The presence of a structured dipolar hydration layer can lead to attractive interactions between like-charged biomacromolecules in solution (and crystal nucleation events). Furthermore, our study suggests that enveloping the protein within a layer of structured solvent (an effect enhanced by IR light) can prevent the protein non-specific aggregation favoring periodic self-assembly. Recognizing the ability to affect protein-water interactions by means of IR radiation may have important implications for biological and bio-inspired systems.

In this paper, an innovative approach for monitoring home electric power quality indicators is presented. Using an electric power analysis device (for this work purpose it will be a smart-meter) and a personal computer it is proved that it is possible to monitor and register electric power quality anomalies, such as long interruptions, voltage dips/swells and frequency oscillations. Through an application developed in Java, a user can view real-time electric parameters, check for electric power quality anomalies and assess load diagram of previous days. Experimental results regarding the application performance are also presented with the respective conclusions.

The present paper aims to examine the potential of the Applied Element Method (AEM) in simulating the blast effects in RC panels. The numerical estimates are compared with the results obtained in an experimental campaign designed to investigate the effectiveness of fibre grout for strengthening full scale RC panels by comparing the effects that a similar blast load produces in a reference and the strengthened panel. First, a numerical model of the reference specimen was created in the software Extreme Loading for Structures and calibrated to match the experimental results. With no further calibration, the fibre reinforced grout strengthening was added and the resulting numerical model subjected to the same blast load. The experimental blast effects on both reference and strengthened panels, despite the lack of high speed measurement equipment (pressure, strains and displacements sensors), compare well with the numerical estimates in terms of residual and maximum displacements, showing that, once calibrated, the AEM numerical models can be successfully used to simulate blast effects in RC panels.

The Late Jurassic Lourinhã Formation is known for its abundant remains of dinosaurs, crocodylomorphs and other vertebrates. Among this record are nine localities that have produced either dinosaur embryos, eggs or eggshell fragments. Herein, we describe and identify the first crocodiloid morphotype eggs and eggshells from the Lourinhã Formation, from five occurrences. One clutch from Cambelas, composed of 13 eggs, eggshell fragments from Casal da Rola and Peralta, one crushed egg and eggshells from Paimogo North, and four crushed eggs as well as eggshell fragments from Paimogo South. We observed and confirmed diagnostic morphological characters for crocodiloid eggshells and which are consistent with a crocodylomorph affinity, such as the ellipsoidal shape, wedge-shaped shell units, triangular extinction under cross-polarized light, and tabular ultrastructure. This material is distinctive enough to propose two new ootaxa within the oofamily Krokolithidae, Suchoolithus portucalensis, oogen. and oosp. nov., for the material from Cambelas, the most complete clutch known for crocodiloid eggs, and Krokolithes dinophilus, oosp. nov., for the remaining material. These are the oldest crocodylomorph eggs known, extending the fossil record for this group to the Late Jurassic. Furthermore, except for the clutch from Cambelas, the material was found with theropod eggs and nests, in the other four occurrences, which seem to suggest some form of biological relationship, still unclear at this point.

The teacher’s knowledge has long been viewed as a strong influence on the students’ learning. Several authors have sought to develop procedures to assess this knowledge, but this has proved to be a complex task. In this paper I present an outline of a conceptualization to analyze the teacher's knowledge, based on the model of the Knowledge for Teaching Mathematics with Technology (KTMT) and a set of tasks. These tasks are chosen by the teacher taking into account the potential of the tasks to take advantage of the technology’s potential. The analysis of the teacher’s KTMT is based on the characteristics of the tasks chosen by the teacher; the balance established between the representations provided by the technology that the tasks advocate; the way how the tasks pay attention to the new issue of seeking for a suitable viewing window; and also the way how the tasks take into account the expectable difficulties of the students in the process of looking for the window.
