The compounds 1-naphthyliminoferrocene{,} 1{,} and 2-methyl-1-naphthyliminoferrocene{,} 2{,} were prepared by the reaction of ferrocenecarboxaldehyde and the corresponding amine{,} 1-naphthylamine and 2-methyl-1-naphthylamine with one drop of pyrrolidine as catalyst{,} in CH2Cl2{,} under inert atmosphere. The compounds were fully characterized by elemental analyses{,} matrix-assisted laser desorption/ionization - time-of-flight mass spectrometry (MALDI-TOF){,} FT-IR{,} 1H and 13C NMR as well as two-dimensional{,} NOESY{,} COSY and HSQC{,} NMR spectroscopy. Mossbauer spectroscopic studies were performed to characterize the iron species in both compounds. The solid-state molecular structures of compounds 1 and 2 were determined by single crystal X-ray diffraction. Analysis of the electrochemical behaviour of complexes 1 and 2 was performed by cyclic and square wave voltammetry. Compounds 1 and 2 were tested as chromogenic and electrochemical sensors for Hg2+.

The non-linear shear behaviour of bovine cortical bone is measured by coupling the Arcan test with digital image correlation. The experimental study is carried out on specimens taken from bovine femur and oriented along the longitudinal-tangential material plane. An ad hoc Arcan fixture is built to transfer the cross-head displacement of the testing machine into a shear loading at the centre of the V-notched section. A validation of predominant shear behaviour at the gauge section is shown from full-field deformation measurements. Moreover, direct evaluation of the shear modulus is obtained by integrating the shear strain component along the V-notches, avoiding numerical correction factors required in the classical data reduction scheme. The shear modulus of bovine cortical bone is found in good agreement with references from literature. Moreover, the shear stress at maximum load is understood as a suitable estimation of the shear strength. Furthermore, the Ramberg�Osgood model is found to provide an accurate description of the non-linear shear behaviour of bone tissue.

Earthquakes, soil settlements, traffic vibrations, and air pollution are some of the actions that can affect historic old buildings. Besides these, the lack of continuous maintenance puts a large part of this heritage in risk due to structural problems that reduce their own safety and that of their users. The preservation and risk mitigation of built cultural heritage require the use of reliable tools in order to assess its state of conservation and to identify and prevent potential vulnerabilities. Having this in mind, it is not possible to carry out destructive tests in most historic old buildings, so it is preferable to opt for nondestructive tests (NDT) or alternative methodologies that allow the physical and mechanical characterization of materials and structure. In this chapter, a general view of NDT methods used in historic buildings to obtain the geometrical information, the damage mapping, the mechanical and physical characterization, and the petrographic analysis of stones is presented. An alternative methodology to physically and mechanically characterize the stone of historic buildings using NDT tests is also proposed. The chapter ends with a case study carried out in the St. Leonard’s Church, a Portuguese monument built in Atouguia da Baleia village in the 13th century, where the alternative methodology here presented was applied. The final results of this study show that the methodology proposed allows the obtention of stress-strain curves in a completely nondestructive way, based on the water absorption coefficient at low pressure.

Abstract The family 81 glycoside hydrolase (GH81) from Clostridium thermocellum is a β-1,3-glucanase belonging to cellulosomal complex. The gene encoding \{GH81\} from Clostridium thermocellum (CtLam81A) was cloned and expressed displaying a molecular mass of  82 kDa. CtLam81A showed maximum activity against laminarin (100 U/mg), followed by curdlan (65 U/mg), at pH 7.0 and 75 °C. CtLam81A displayed Km, 2.1 ± 0.12 mg/ml and Vmax, 109 ± 1.8 U/mg, against laminarin under optimized conditions. CtLam81A activity was significantly enhanced by Ca2+ or Mg2+ ions. Melting curve analysis of CtLam81A showed an increase in melting temperature from 91 °C to 96 °C by Ca2+ or Mg2+ ions and decreased to 82 °C by EDTA, indicating that Ca2+ and Mg2+ ions may be involved in catalysis and in maintaining structural integrity. \{TLC\} and MALDI-TOF analysis of β-1,3-glucan hydrolysed products released initially, showed β-1,3-glucan-oligosaccharides degree of polymerization (DP) from \{DP2\} to DP7, confirming an endo-mode of action. The catalytically inactive mutant CtLam81A-E515A generated by site-directed mutagenesis was co-crystallized and tetragonal crystals diffracting up to 1.4 Å resolution were obtained. CtLam81A-E515A contained 15 α-helices and 38 β-strands forming a four-domain structure viz. a β-sandwich domain I at N-terminal, an α/β-domain II, an (α/α)6 barrel domain III, and a small 5-stranded β-sandwich domain IV.

This work addresses the reconstruction of strain gradient fields at the wood growth ring scale from full-field deformation measurements provided by digital image correlation. Moreover, the spatial distribution of the earlywood and latewood radial modulus of elasticity is assessed. Meso-scale tensile tests are carried out on Pinus pinaster Ait. wooden specimens oriented in the radial–tangential plane under quasi-static loading conditions. A parametric analysis of the two-dimensional digital image correlation extrinsic and intrinsic setting parameters is performed, in a balance between spatial resolution and resolution. It is shown that the parametric module is an effective way to quantitatively support the choice of digital image correlation parameters in the presence of the high deformation gradient fields generated by the structure–property relationships at the scale of observation. Under the assumption of a uniaxial tensile stress state, the spatial distribution of the radial elastic modulus across the growth rings is obtained. It is observed that the ratio of the radial modulus of elasticity between latewood and earlywood tissues can vary significantly as a function of the digital image correlation parameters. It is pointed out, however, that a convergence value can be systematically established. Effectively, earlywood and latewood stress–strain curves are obtained and elastic properties are determined assuming the converged digital image correlation setting parameters.

Abstract The authors have recently proposed an innovative connector system that consists on a Glass Fibre Reinforced Polymer (GFRP) perforated plate that is embedded into Steel Fibre Reinforced Self-Compacting Concrete (SFRSCC) layers. The connection is strongly based in the mechanical interlock assured by the dowels originated from the \{SFRSCC\} passing through the holes opened on the \{GFRP\} plates. In this study, an analytical framework to evaluate the load capacity of the connections when loaded transversally was developed based on experimental pull-out tests presented in the companion paper (Part I). For a better understanding of the mechanical behaviour of the connections and to allow to make estimations of the load capacity of connection when it is conditioned by the rupture of the connector itself, pull-out pin-bearing tests with single-hole plates were executed to assess the effect of the type of \{GFRP\} on the strain distribution in the vicinity of the holes until the failure, as well as the estimated failure modes and load capacities of the connections.
