A aprendizagem de leitura implica no desenvolvimento de competências e habilidades e as TICs possuem o papel de gerar estratégias de relação com o saber. Esta pesquisa identifica competências para uma maior autonomia do sujeito em leitura, via relação entre as tecnologias da informação e comunicação e outros letramentos, através da investigação sobre a prática pedagógica de letramento digital na escola fundamental, no sétimo ano. O método consiste em um estudo, de caráter quantitativo, no qual são analisados os contextos de uso do letramento digital, a junção de diversos outros letramentos e habilidades subjacentes ao letramento digital e a interrelação com plataformas (de gestão de aprendizagem, imagens e vídeos), tecnologias para publicação e partilha de conteúdos, tecnologias de colaboração e construção, redes sociais e tecnologias de comunicação interpessoal. Depois, uma abordagem complementar de investigação-ação Design-Based Research (Db-R) estuda de que forma deve o professor trabalhar com os alunos para favorecer essa autonomia. Os resultados apontam para um conjunto de ações, nas políticas públicas nas práticas pedagógicas, com medidas a superar as dificuldades e os atavismos, com a inserção dos estudantes em práticas com letramento digital e emprego de ferramentas e recursos desenvolvidos de modo específico para o público final. Pseudo-cyclic and cyclic loading were applied to CFRP-to-steel bonded joints built with two different CFRP laminates. In this paper, the strength capacity and bond-slip curves are presented and compared. The modes of failure are also described and associated with the types of material used, and the observed performances are correlated. The analysis of the results showed a threshold value for loading and amplitude level, below which the cyclic loading caused no detectable damage. For cycles above that limit, the region of the joints around the loaded end presented degradation reflected on the bond-slip stiffness and on the increase of residual deformation. It was found that the normalized dissipated energies either obtained from the bond-slip relationship or from the load-slip response had the same trend. The experimental data allowed also to establish a relationship between the damage developed within the interface and the normalized slip. A preliminary estimate of fatigue limit based on those data is suggested. © 2018 Elsevier Ltd

The p53 tumor suppressor is widely found to be mutated in human cancer. This protein is regarded as a molecular hub regulating different cell responses, namely cell death. Compelling data have demonstrated that the impairment of p53 activity correlates with tumor development and maintenance. For these reasons, the reactivation of p53 function is regarded as a promising strategy to halt cancer. In the present work, the recombinant mutant p53R280K DNA binding domain (DBD) was produced for the first time, and its crystal structure was determined in the absence of DNA to a resolution of 2.0 Å. The solved structure contains four molecules in the asymmetric unit, four zinc(II) ions, and 336 water molecules. The structure was compared with the wild-type p53 DBD structure, isolated and in complex with DNA. These comparisons contributed to a deeper understanding of the mutant p53R280K structure, as well as the loss of DNA binding related to halted transcriptional activity. The structural information derived may also contribute to the rational design of mutant p53 reactivating molecules with potential application in cancer treatment.

The compounds 1-naphthyliminoferrocene{,} 1{,} and 2-methyl-1-naphthyliminoferrocene{,} 2{,} were prepared by the reaction of ferrocenecarboxaldehyde and the corresponding amine{,} 1-naphthylamine and 2-methyl-1-naphthylamine with one drop of pyrrolidine as catalyst{,} in CH2Cl2{,} under inert atmosphere. The compounds were fully characterized by elemental analyses{,} matrix-assisted laser desorption/ionization - time-of-flight mass spectrometry (MALDI-TOF){,} FT-IR{,} 1H and 13C NMR as well as two-dimensional{,} NOESY{,} COSY and HSQC{,} NMR spectroscopy. Mossbauer spectroscopic studies were performed to characterize the iron species in both compounds. The solid-state molecular structures of compounds 1 and 2 were determined by single crystal X-ray diffraction. Analysis of the electrochemical behaviour of complexes 1 and 2 was performed by cyclic and square wave voltammetry. Compounds 1 and 2 were tested as chromogenic and electrochemical sensors for Hg2+.

Abstract The family 81 glycoside hydrolase (GH81) from Clostridium thermocellum is a β-1,3-glucanase belonging to cellulosomal complex. The gene encoding \{GH81\} from Clostridium thermocellum (CtLam81A) was cloned and expressed displaying a molecular mass of  82 kDa. CtLam81A showed maximum activity against laminarin (100 U/mg), followed by curdlan (65 U/mg), at pH 7.0 and 75 °C. CtLam81A displayed Km, 2.1 ± 0.12 mg/ml and Vmax, 109 ± 1.8 U/mg, against laminarin under optimized conditions. CtLam81A activity was significantly enhanced by Ca2+ or Mg2+ ions. Melting curve analysis of CtLam81A showed an increase in melting temperature from 91 °C to 96 °C by Ca2+ or Mg2+ ions and decreased to 82 °C by EDTA, indicating that Ca2+ and Mg2+ ions may be involved in catalysis and in maintaining structural integrity. \{TLC\} and MALDI-TOF analysis of β-1,3-glucan hydrolysed products released initially, showed β-1,3-glucan-oligosaccharides degree of polymerization (DP) from \{DP2\} to DP7, confirming an endo-mode of action. The catalytically inactive mutant CtLam81A-E515A generated by site-directed mutagenesis was co-crystallized and tetragonal crystals diffracting up to 1.4 Å resolution were obtained. CtLam81A-E515A contained 15 α-helices and 38 β-strands forming a four-domain structure viz. a β-sandwich domain I at N-terminal, an α/β-domain II, an (α/α)6 barrel domain III, and a small 5-stranded β-sandwich domain IV.
