Main conclusion: The phenylpropanoid pathway impacts the cork quality development. In cork of bad quality, the flavonoid route is favored, whereas in good quality, cork lignin and suberin production prevails. Cork oaks develop a thick cork tissue as a protective shield that results of the continuous activity of a secondary meristem, the cork cambium, or phellogen. Most studies applied to developmental processes do not consider the cell types from which the samples were extracted. Here, laser microdissection (LM) coupled with transcript profiling using RNA sequencing (454 pyrosequencing) was applied to phellogen cells of trees producing low- and good quality cork. Functional annotation and functional enrichment analyses showed that stress-related genes are enriched in samples extracted from trees producing good quality cork (GQC). This process is under tight transcriptional (transcription factors, kinases) regulation and also hormonal control involving ABA, ethylene, and auxins. The phellogen cells collected from trees producing bad quality cork (BQC) show a consistent up-regulation of genes belonging to the flavonoid pathway as a response to stress. They also display a different modulation of cell wall genes resulting into a thinner cork layer, i.e., less meristematic activity. Based on the analysis of the phenylpropanoid pathway regulating genes, in GQC, the synthesis of lignin and suberin is promoted, whereas in BQC, the same pathway favors the biosynthesis of free phenolic compounds. This study provided new insights of how cell-specific gene expression can determine tissue and organ morphology and physiology and identified robust candidate genes that can be used in breeding programs aiming at improving cork quality.

In this work, we highlight the influence of three different sol stabilizers, namely diethanolamine (DEA), Ammonium Hydroxide (NH4OH), and Nitric Acid (HNO3), on the optical and structural properties of spin-coated zinc oxide (ZnO) thin films. The XRD patterns related to all films exhibit a hexagonal crystal structure with a preferential orientation along the (0 0 2) direction. However an additional {\textless}100{\textgreater} peak arises when the films are prepared with DEA and NH4OH showing a better crystallinity than that displayed by HNO3-prepared films. The elaborated films show a high transparency reaching 80{%} for DEA-prepared films. The analysis of the transmittance and the reflectance measurements confirms a direct band-to-band transition. Depending on the sol stabilizer, the optical band gap energy is varying from 3.16 to 3.22 eV. The relatively wide band-gap of DEA-prepared ZnO films is correlated to their high crystallinity. Room temperature photoluminescence spectra indicate strong UV emission at around 377 nm originated from nearby band-edge transitions. Yet, the use of DEA as a stabilizer leads to a net intensity increase of the blue peak emission.

The high global burden of over one million annual lethal fungal infections reflects a lack of protective vaccines, late diagnosis and inadequate chemotherapy. Here, we have generated a unique set of fully human anti-Candida monoclonal antibodies (mAbs) with diagnostic and therapeutic potential by expressing recombinant antibodies from genes cloned from the B cells of patients suffering from candidiasis. Single class switched memory B cells isolated from donors serum-positive for anti-Candida IgG were differentiated in vitro and screened against recombinant Candida albicans Hyr1 cell wall protein and whole fungal cell wall preparations. Antibody genes from Candida-reactive B cell cultures were cloned and expressed in Expi293F human embryonic kidney cells to generate a panel of human recombinant anti-Candida mAbs that demonstrate morphology-specific, high avidity binding to the cell wall. The species-specific and pan-Candida mAbs generated through this technology display favourable properties for diagnostics, strong opsono-phagocytic activity of macrophages in vitro, and protection in a murine model of disseminated candidiasis.

Semiconductor nanowires are mostly processed by complex, expensive and high temperature methods. In this work, with the intent of developing zinc tin oxide nanowires (ZTO NWs) by low-cost and low-complexity processes, we show a detailed study on the influence of chemical parameters in the hydrothermal synthesis of ZTO nanostructures at temperatures of only 200 °C. Two different zinc precursors, the ratio between zinc and tin precursors, the concentration of the surfactant agent and of the mineralizer were studied. The type and the crystallinity of the nanostructures was found to be highly dependent on the used precursors and on the concentration of each reagent. Conditions for obtaining different ZTO nanostructures were achieved, namely Zn2SnO4 nanoparticles and ZnSnO3 nanowires with length ≈ 600 nm, with the latter being reported for the first time ever by hydrothermal methods without the use of seed layers. This article presents a nonlinear analytical solution for the prediction of the full-range debonding response of mechanically anchored, fiber-reinforced polymer (FRP) composites from the substrate. The nonlinear analytical approach predicts, for any monotonic loading history or bonded length, the relative displacements (or slips) between materials, the strains in the FRP composite, the bond stresses within the interface, and the stresses developed in the substrate. The load-slip responses of FRP-to-substrate interfaces with short and long bonded lengths are motives of analysis and discussion. The solutions obtained from the proposed approach are also compared with other experimental results found in the literature.

Additive manufacturing (commonly known as 3D printing or rapid prototyping) refers to processes used to produce parts in an additive manner by means of computer-aided design (CAD). While additive manufacturing is a technology that can be used in many different application areas, this project focuses on future trends in additive manufacturing (AM) aimed at improving biological functionality (bio-AM) and on its opportunities, barriers and challenges. The big advantage of this technique is that small batches can be produced more economically than with any other manufacturing process. Virtually any structure can be customized, which is particularly important in the healthcare sector. Possible applications include biological implants such as organs and tissues, nutrients, drugs and their transport mechanisms, equipment such as surgical knives and drilling guides, tissues for research, development and training, and personalized prostheses, supports and exoskeletons. Besides exploring such applications, the project will also systematically analyze potential "human enhancement" uses of AM technology and developments in the emerging do-it-yourself (DIY) cultures ("bio/body-hacking"; cyborgism; open source 3D printing movement).

In the first phase of the project, the technological state of the art will be analyzed, as will a wide variety of non-technical aspects, regulatory issues and future trends, also with a special focus on sociotechnical imaginaries (e.g., in science fiction), human enhancement and DIY cultures. This horizon scanning will be accomplished partly by means of expert and stakeholder interviews.

In the second phase, the project will use a variety of foresight and technology assessment methods and will carry out a 360° envisioning exercise with contributions by external experts, entailing an in-depth analysis of selected applications of bio-AM.

The project work will end with a scenario development phase in which the focus will be on likely outcomes of already emerging developments, though further-reaching future perspectives will be taken into account to a certain extent. Taken together, these scenarios will allow for both a broader understanding of the wide range of potential impacts of AM applications and a clearer picture of potential policy challenges relevant to the Members of the European Parliament.

Cellulosomes are highly sophisticated molecular nanomachines that participate in the deconstruction of complex polysaccharides, notably cellulose and hemicellulose. Cellulosomal assembly is orchestrated by the interaction of enzyme-borne dockerin (Doc) modules to tandem cohesin (Coh) modules of a non-catalytic primary scaffoldin. In some cases, as exemplified by the cellulosome of the major cellulolytic ruminal bacterium Ruminococcus flavefaciens, primary scaffoldins bind to adaptor scaffoldins that further interact with the cell surface via anchoring scaffoldins, thereby increasing cellulosome complexity. Here we elucidate the structure of the unique Doc of R. flavefaciens FD-1 primary scaffoldin ScaA, bound to Coh 5 of the adaptor scaffoldin ScaB. The RfCohScaB5-DocScaA complex has an elliptical architecture similar to previously described complexes from a variety of ecological niches. ScaA Doc presents a single-binding mode, analogous to that described for the other two Coh-Doc specificities required for cellulosome assembly in R. flavefaciens. The exclusive reliance on a single-mode of Coh recognition contrasts with the majority of cellulosomes from other bacterial species described to date, where Docs contain two similar Coh-binding interfaces promoting a dual-binding mode. The discrete Coh-Doc interactions observed in ruminal cellulosomes suggest an adaptation to the exquisite properties of the rumen environment.

This paper presents a study of the behaviour and load capacity of Steel Fibre Reinforced Concrete (SFRC) flat slabs under monotonically increased concentrated vertical loads. The SFRC was used only in the local region of the slab-column connection, as the rest of the slab was cast using normal concrete (NC) without fibres. The six experimental test specimens had a thickness of 150 mm with different longitudinal reinforcement ratios, using a non-uniform distribution over the slab width. The concretes used were made with different Dramix® 4D 65/60 BG steel fibre contents (0%, 0.5 %, 0.75% and 1.0% volume content). The slab tests were complemented by flexural tests on notched-beams. This made it possible to determine the tension behaviour of the different concretes used, through a linear post-cracking behavior and inverse analysis. The inverse analysis made it possible to define the stress-crack opening relationship that characterize the tension behaviour of SFRC and to relate it to the observed behaviour and load capacity of the tested slabs. The tests results show that the tensile behaviour of the SFRC plays an important role in the behavioural and load capacity of the slabs and that it can be considered relevant to physically based models.
