Respiratory nitric oxide reductase (NOR) was purified from membrane extract of Pseudomonas (Ps.) nautica cells to homogeneity as judged by polyacrylamide gel electrophoresis. The purified protein is a heterodimer with subunits of molecular masses of 54 and 18 kDa. The gene encoding both subunits was cloned and sequenced. The amino acid sequence shows strong homology with enzymes of the cNOR class. Iron/heme determinations show that one heme c is present in the small subunit (NORC) and that approximately two heme b and one non-heme iron are associated with the large subunit (NORB), in agreement with the available data for enzymes of the cNOR class. Mossbauer characterization of the as-purified, ascorbate-reduced, and dithionite-reduced enzyme confirms the presence of three heme groups (the catalytic heme b(3) and the electron transfer heme b and heme c) and one redox-active non-heme Fe (Fe-B). Consistent with results obtained for other cNORs, heme c and heme b in Ps. nautica cNOR were found to be low-spin while FeB was found to be high-spin. Unexpectedly, as opposed to the presumed high-spin state for heme b(3), the Mossbauer data demonstrate unambiguously that heme b(3) is, in fact, low-spin in both ferric and ferrous states, suggesting that heme b(3) is six-coordinated regardless of its oxidation state. EPR spectroscopic measurements of the as-purified enzyme show resonances at the g similar to 6 and g similar to 2-3 regions very similar to those reported previously for other cNORs. The signals at g = 3.60, 2.99, 2.26, and 1.43 are attributed to the two charge-transfer low-spin ferric heme c and heme b. Previously, resonances at the g similar to 6 region were assigned to a small quantity of uncoupled high-spin Fe-III heme b(3). This assignment is now questionable because heme b(3) is low-spin. On the basis of our spectroscopic data, we argue that the g = 6.34 signal is likely arising from a spin spin coupled binuclear center comprising the low-spin Fe-III heme b(3) and the high-spin Fe-B(III). Activity assays performed under various reducing conditions indicate that heme b(3) has to be reduced for the enzyme to be active. But, from an energetic point of view, the formation of a ferrous heme-NO as an initial reaction intermediate for NO reduction is disfavored because heme [FeNO](7) is a stable product. We suspect that the presence of a sixth ligand in the Fe-II-heme b(3) may weaken its affinity for NO and thus promotes, in the first catalytic step, binding of NO at the Fe-B(II) site. The function of heme b(3) would then be to orient the Fe-B-bound NO molecules for the formation of the N-N bond and to provide reducing equivalents for NO reduction.

We have investigated the electrical properties of metal-semiconductor field-effect transistors (MESFET) based on amorphous oxide semiconductor channels. All functional parts of the devices were sputter-deposited at room temperature. The influence on the electrical properties of a 150 °C annealing step of the gallium-indium-zinc-oxide channel is investigated. The MESFET technology offers a simple route for processing of the transistors with excellent electrical properties such as low subthreshold swing of 112 mV/decade, gate sweep voltages of 2.5 V, and channel mobilities up to 15 cm2 /V s. © 2010 American Institute of Physics.

This paper discusses the properties of sputtered multicomponent amorphous dielectrics based on mixtures of high-κ and high-bandgap materials and their integration in oxide TFTs, with processing temperatures not exceeding 1 50° C. Even if Ta2O5 films are already amorphous, multicomponent materials such as Ta2O5-SiO2 and Ta2O5-Al2O3 allow an increase in the bandgap and the smoothness of the films, reducing their leakage current and improving (in the case of Ta2O5-SiO2)the dielectric/semiconductor interface properties when these dielectrics are integrated in TFTs. For HfO2- based dielectrics, the advantages of multicomponent materials are even clearer: while HfO2 films present a polycrystalline structure and a rough surface, HfO2-SiO2 films exhibit an amorphous structure and a very smooth surface. The integration of the multicomponent dielectrics in GIZO TFTs allows remarkable performance, comparable with that of GIZO TFTs using SiO2 deposited at 400°C by PECVD. For instance, with Ta2O5-SiO2 as the dielectric layer, field-effect mobility of 35 cm2/(V-sec), close to 0 V turn-on voltage, an on/off ratio higher than 106, a subthreshold slope of 0.24 V/dec, and a small/recoverable threshold voltage shifts under constant current (ID = 10 μ,A) stress during 24 hours are achieved. Initial results with multilayers of SiO2/HfO 2-SiO2/SiO2 are also shown, allowing a lower leakage current with lower thickness and excellent device performance. © Copyright 2010 Society for Information Display.

A ligação pilar-fundação tem um papel fundamental no comportamento das estruturas pré-fabricadas, em particular, na presença de acções horizontais, como a acção dos sismos e do vento, que introduzem esforços elevados na base dos pilares. Um dos sistemas de ligação pilar-fundação usados em estruturas pré-fabricadas consiste em deixar na extremidade inferior dos pilares armaduras salientes que são introduzidas em negativos executados nas fundações. Estes negativos são posteriormente injectados com grout para garantir a ligação. No presente trabalho foram estudadas diferentes soluções de negativos para ligação das armaduras do pilar à fundação, tendo sido realizados ensaios monotónicos para analisar o seu comportamento. Nesta comunicação apresentam-se e discutem-se os resultados obtidos da análise experimental efectuada.

Estes negativos são posteriormente injectados com “grout” para garantir a ligação. Foram estudados diferentes soluções de negativos para ligação das armaduras do pilar na fundação. Foram realizados ensaios monotónicos e cíclicos, para analisar não só o comportamento às acções estáticas mas também para a acção sísmica. No presente trabalho apresentam-se os resultados obtidos com a análise experimental efectuada. O estudo realizado permite apresentar recomendações para projecto e quais os tipos de ligações com melhor desempenho.

Portugal has been providing dinosaur remains since, at least, 1863. The 18th century tiles depicting the legend of Our Lady in Cabo Espichel are probably the oldest known dinosaur track illustration. To our knowledge, the first remains found in Portugal were theropod teeth collected near Porto das Barcas (Late Jurassic of Lourinhã) in June 20th, 1863 by the geologist Carlos Ribeiro (1813-1882). The first dinosaur paper was written by Henri Sauvage (1842-1917) published in 1896. All remains collected since 19th century were gathered in a work signed by Albert de Lapparent (1905-1975) and Georges Zbyszewski (1909-1999 ) titled Les Dinosauriens du Portugal (1957) that was a significant milestone in the Portuguese dinosaur paleontology and gives the state-of-the-art by the time. Several dinosaurs are named, described, depicted and mapped in that monograph. The first track record is given by Jacinto Pedro Gomes (1844-1916) in 1916. Concerning the non-scientific literature referring to dinosaurs, in 1884 the newspaper Occidente reports the Bernissart findings in Belgium. In the 1959 occurs the first visit to Portugal of Walter Kühne (1911-1991) from the Free University of Berlin. Further visits and work granted the access to the Guimarota Mine and other Late Jurassic deposits in the 1960’s, 70’s and 80’s with a high number of publications. In the 1980’s and early 1990’s starts a progressive era for dinosaur paleontology in Portugal with the works of Peter Galton, Miguel Telles Antunes, the Natural History Museum, the Museum of Lourinhã and the New University of Lisbon, Oliver Rauhut, and others.
