The final step of bacterial denitrification, the two-electron reduction of N2O to N-2, is catalyzed by a multi-copper enzyme named nitrous oxide reductase. The catalytic centre of this enzyme is a tetranuclear copper site called CuZ, unique in biological systems. The in vitro reconstruction of the activity requires a slow activation in the presence of the artificial electron donor, reduced methyl viologen, necessary to reduce CuZ from the resting non-active state (1Cu(II)/3Cu(I)) to the fully reduced state (4Cu(I)), in contrast to the turnover cycle, which is very fast. In the present work, the direct reaction of the activated form of Pseudomonas nautica nitrous oxide reductase with stoichiometric amounts of N2O allowed the identification of a new reactive intermediate of the catalytic centre, CuZA degrees, in the turnover cycle, characterized by an intense absorption band at 680 nm. Moreover, the first mediated electrochemical study of Ps. nautica nitrous oxide reductase with its physiological electron donor, cytochrome c-552, was performed. The intermolecular electron transfer was analysed by cyclic voltammetry, under catalytic conditions, and a second-order rate constant of (5.5 +/- A 0.9) x 10(5) M-1 s(-1) was determined. Both the reaction of stoichiometric amounts of substrate and the electrochemical studies show that the active CuZA degrees species, generated in the absence of reductants, can rearrange to the resting non-active CuZ state. In this light, new aspects of the catalytic and activation/inactivation mechanism of the enzyme are discussed.

Models, with different levels of detail, share similar abstractions that can be reused by means of model-driven techniques such as transformations. For example, scenarios are a well-known technique in requirements engineering to represent behavioral flows in a software system. When using UML, scenarios are typically represented with activity models in the early stages of software development, while sequence models are used to describe more detailed object interactions as modeling progresses. This paper defines transformation rules to automate the migration from activity to sequence models. We present a case study illustrating the application of our transformation rules. Our preliminary assessment of the impact of the benefits of using these transformations points to: (i) a reduction of around 50% in the effort building sequence models, (ii) increased trace ability among models, and (iii) error prevention when migrating from different scenario notations.

The Upper Jurassic of Portugal has a rich vertebrate fauna well documented from both body and trace fossils. Although the occurrence of crocodyles and pterosaurs is well documented from body fossils, trace fossils from both groups were unknown until now. Here we describe an isolated crocodyle-like track from Praia da Peralta and pterosaur tracks from the Kimmeridgian of Pedreira do Avelino, Sesimbra (Azóia Fm.) and Porto das Barcas, Lourinhã (Lourinhã Fm.). An enigmatic track suggests the possible presence of a small, tail-dragging tetrapod. Possible track-makers are suggested based on the known Late Jurassic vertebrate fauna of Portugal.
