Perante as conhecidas dificuldades em alcançar uma adequada integração da tecnologia no processo de ensino e aprendizagem da Matemática, este estudo pretende, apoiando-se na formação ao nível da tecnologia ministrada em duas instituições europeias, identificar aspetos com potencial para promover a formação inicial, no âmbito da tecnologia, de professores de Matemática. Adota-se uma metodologia de índole qualitativa e interpretativa, sendo os dados recolhidos de natureza documental ou relativos a trabalhos de análise e reflexão crítica realizados por dois futuros professores (um de cada instituição). As principais conclusões alcançadas apontam para grandes diferenças entre os contextos de formação, com uma das instituições a valorizar de forma mais significativa a formação na área. Ainda assim, os futuros professores de ambas as instituições mostram alguma tendência para escolher tarefas onde a exploração que é feita da tecnologia fica aquém do seu potencial, onde o recurso ao papel e lápis está sempre presente, e onde a reflexão em torno das características das tarefas e da sua implementação parece ser algo superficial. Apesar da complexidade do processo de integração da tecnologia nas práticas, os aspetos referidos parecem-nos ser dignos de atenção em qualquer programa de formação inicial de professores de Matemática.

Acrylamide is a neurotoxic and carcinogenic organic compound that is able to bind to several biomolecules and form adducts, through nucleophilic addition and in vivo by the Maillard Reaction, interfering with the biological functions of these molecules. Hemoglobin is one of the most abundant intracellular blood proteins, and thus it is of high interest to understand whether the binding of acrylamide can alter its properties. The interaction of acrylamide with hemoglobin was assessed in a 20:1 ratio, and after a 72 h-incubation period, a decrease of ca. 50% in the absorbance of the hemoglobin's Soret band was observed at 37 °C. This together with the analysis of circular dichroism spectra indicate that acrylamide binds in close proximity to the heme group. These perturbations were confirmed to not correspond to the loss of the heme group and were mostly reverted after passing the protein through a size-exclusion chromatographic matrix, suggesting a dominant non-covalent interaction for the observed effect. The thermodynamic parameters of unfolding in the absence and presence of acrylamide, suggest an interaction based on H-bonds and van der Waals forces that slightly stabilizes hemoglobin. The oxygen binding capacity of hemoglobin does not seem to be hindered, as no differences in the Q bands were observed in the adduct.

BackgroundHalf of human cancers harbour TP53 mutations that render p53 inactive as a tumor suppressor. As such, reactivation of mutant (mut)p53 through restoration of wild-type (wt)-like function represents one of the most promising therapeutic strategies in cancer treatment. Recently, we have reported the (S)-tryptophanol-derived oxazoloisoindolinone SLMP53-1 as a new reactivator of wt and mutp53 R280K with in vitro and in vivo p53-dependent antitumor activity. The present work aimed a mechanistic elucidation of mutp53 reactivation by SLMP53-1.
Methods and results
By cellular thermal shift assay (CETSA), it is shown that SLMP53-1 induces wt and mutp53 R280K thermal stabilization, which is indicative of intermolecular interactions with these proteins. Accordingly, in silico studies of wt and mutp53 R280K DNA-binding domain with SLMP53-1 unveiled that the compound binds at the interface of the p53 homodimer with the DNA minor groove. Additionally, using yeast and p53-null tumor cells ectopically expressing distinct highly prevalent mutp53, the ability of SLMP53-1 to reactivate multiple mutp53 is evidenced.
Conclusions
SLMP53-1 is a p53-activating agent with the ability to directly target wt and a set of hotspot mutp53.
General Significance
This work reinforces the encouraging application of SLMP53-1 in the personalized treatment of cancer patients harboring distinct p53 status.

Mathematics has a strong presence in the school curriculum, often justified by its usefulness in social life, in the world of work and by its connections with other sciences. This interdisciplinary connection, in particular when it requires constructing and refining mathematical models and discussing their applications to solve problems of other sciences, can assist students to understand why mathematics is so important in school. In the development of interdisciplinary activities, the characteristics of the tasks emerge as an important aspect. The emphasis is on the use of technological materials and the way they can support the development of concepts, provide different representations and support deeper understandings, and offer a multifaceted support to collect data and simulate experiences. Based on these assumptions, the aim of this chapter is to present, analyse and discuss tasks that promote interdisciplinary technological approaches from a mathematical point of view. In this chapter we assume interdisciplinarity as a complex construct, and in order to clarify its meaning we will discuss several types of conceptions, from multidisciplinary, to interdisciplinary, and to transdisciplinary. We will then address related concepts, such as modelling and STEM, highlighting similarities and differences between them, to reach an understanding of interdisciplinarity. In the process of the interdiciplinary approach, digital technologies arise as a central element. Based on a set of tasks on mathematics and on different sciences, we discuss what can change on an interdisciplinary approach to the teaching and learning of mathematical content and on the articulation between subjects.

This paper describes the experimental campaign to study the behaviour of reinforced-concrete flat slab structures with steel stirrups as punching shear reinforcement, under combined vertical and horizontal cyclic loading. The vertical load was first applied and kept constant during the test, while, regarding the cyclic horizontal loading, imposed cyclic drifts were increased until failure. Four slab specimens with shear reinforcement were tested and the results compared to a control slab specimen without shear reinforcement. The studied variables were different shear reinforcement ratios and the number of stirrup layers. The slabs were 4·15 × 1·85 m2 and 0·15 m thick, connected to two steel half-columns. The test setup used was developed by the research team and aimed to simulate the boundary conditions of a flat slab, representing the slab between middle spans in one direction and between zero bending moment points in the other direction. Results show that the use of steel stirrups as shear reinforcement is very effective, increasing shear, drift and energy dissipation capacities. The obtained results were also compared to the provisions given by European and American codes.

The paper is focused on the calibration of non-linear 3D finite element (FE) analyses to simulate punching failure of R/C flat slabs. The calibration procedure is developed with reference to the code ABAQUS, which is one of the most used computer codes in nonlinear modelling of R/C structures. Generally, the calibration of a nonlinear FE model is grounded on one test only, so its reliability could be limited. Here a hybrid method for the calibration of FE models of R/C flat slabs failing in punching is proposed and discussed. The method consists in calibrating input data by comparison of finite element model (FEM) results with both experimental data and predictions provided by analytical models. The procedure allows for a consistent calibration to be performed, valid for a wide range of longitudinal reinforcement ratios, from 0.5% to 2.00%, and concrete grades, from C20/25 to C50/60. A case study is investigated using the proposed method. Results show that calibrated values of the fracture energy lie between those provided by Model Code 1990 and Model Code 2010. From the new calibration procedure, a relationship between fracture energy and concrete compressive strength is also derived and blind analyses are performed to check its reliability against experimental results.

This preliminary report presents the outcomes of desk research, which was oriented to obtain a clear picture of the ‘state of the art’ in the involved countries and sectors. The report first reviews the main concepts and categories used to approach the topic and provides a definition of virtual work. Second, it describes the incidence and features of virtual work based on both European and national surveys. Third, it reviews previous research outcomes on the effects of virtual work on working conditions at national and sectoral level. Fourth, it analyses national approach to regulate virtual work. Fifth, the report addresses main institutional features of industrial relations in the three sectors and five countries; and the role played by social dialogue and collective bargaining at sectoral and company level to regulate working conditions of virtual workers.
