Viral proteins typically exist in the context of complex virions or in the even more complex host cells in which they replicate. Hence, meaningful insights into virus protein structure often need to account for this context. Various flavors of in situ cryo-electron microscopy (cryo-EM), such as cryo-electron tomography, are key methods for the contextual study of virus protein structure in pleomorphic virions and host cells. Here, we review recent in situ cryo-EM work on three selected phenomena in RNA virus replication: the maturation and nuclear entry of HIV-1, the membrane-bound replication organelles of positive-sense RNA viruses, and the membrane-less viral factories of negative-sense RNA viruses. We highlight cases where the imaged phenomena are targets of novel antiviral drugs (such as the recently approved antiretroviral Lenacapavir), drug candidates, and antiviral strategies. Finally, we discuss recent technical advances that extend the reach of in situ cryo-EM in virology.

Accurate blast models are essential for disaster management and emergency preparedness. Semi-empirical methods, which rely on free-field assumptions, struggle to provide precise data for complex building shapes because they ignore shock wave reflections. This research numerically investigates the effects of an explosion on a large building of complex geometrical configuration using blastFoam, estimating the façade damage and assessing risks to occupants. To evaluate the importance of accurately modelling the building’s geometry, four levels of detail were considered. Additionally, simulations were also performed using the Load Blast Enhanced (LBE) method from LS-DYNA to compare the estimates of this faster semi-empirical approach with those obtained through CFD analyses. The findings reveal that simpler CFD models are adequate for façade analysis and injury assessment around the building but fall short for predicting injury distribution within interior spaces or between buildings. The LBE underestimates both structural damage and human injury levels, while detailed CFD highlights the importance of accounting for interior walls and windows to enhance blast pressure predictions.

Open-cell foams based on hydroxyapatite (HAp) can mimic the extracellular matrix (ECM) to better replace damaged hard tissues and assist in their regeneration processes. Aerogels of HAp nanowires (NW) with barium titanate (BT) particles were produced and characterized regarding their physical and chemical properties, bioactivity, and in vitro cytotoxicity. Considering the role of piezoelectricity (mainly due to collagen) and surface charges in bone remodeling, all BT particles, of size 280 nm and 2 and 3 µm, contained BaTiO3 in their piezoelectric tetragonal phase. The synthesized nanowires were verified to be AB-type carbonated hydroxyapatite. The aerogels showed high porosity and relatively homogeneous distribution of the BT particles. Barium titanate proved to be non-cytotoxic while all the aerogels produced were cytotoxic for an extract concentration of 1 mg/mL but became non-cytotoxic at concentrations of 0.5 mg/mL and below. It is possible that these results were affected by the higher surface area and quicker dissolution rate of the aerogels. In the bioactivity assays, SEM/EDS, it was not easy to differentiate between the apatite deposition and the surface of the HAp wires. However, a quantitative EDS analysis shows a possible CaP deposition/dissolution cycle taking place.

Abstract The Monastery of Alcobaça houses in reserve the ceramic tiles that adorned the floor of the church's apse. These tiles were removed during rehabilitation works and many preserve part of their original fixing mortars. A comprehensive analysis of 21 samples was conducted using a multi-analytical approach (X-ray fluorescence, X-ray diffraction, scanning electron microscopy–energy-dispersive X-ray spectroscopy, μ-Raman spectroscopy, thermogravimetry–differential thermal analysis, optical microscopy, and colorimetry). Results suggest compositional variations in the samples from the back and sides of the ceramic tiles; however, the mineralogy and general characteristics of the aggregates remained consistent between the samples and are coherent with the local geology, suggesting a shared historical origin.

Political and economic crises and issues such as global warming, decreased fossil fuel reserves, environmental pollutions and acid rains are just some of the problems caused by improper energy consumption. Energy efficiency optimization with the right policy and definition of accurate scientific methods proven in energy consumption includes planning, enforcement, organizing permanent control, and continuously reviewing technology upgrades. Additionally, developing legal solutions to implement these programs prevents the destruction and waste of energy resources and reduces the side effects of improper energy consumption. The shape of the city and its components can affect the city’s quality and its spaces, as well as the air quality change, creating a particular climate in their area, causing phenomena such as heat islands, dust, rain, albedo, atmospheric inversion, and some others. By making changes in the shape of the city, both on a large scale and a micro-scale, atmospheric currents and, consequently, air quality in cities can be improved. Urban morphological indicators, including building height, building type, occupancy level, segmentation, block size, block shape, density, and urban green, can affect the weather and climate conditions outside and inside the building and, thus, the energy demand in the areas. By creating effective reforms in the model of configuration of the urban regions, including the pattern of land fragmentation, the physical characteristics of buildings, the network of passages and open spaces, it is possible to increase the energy efficiency of buildings in the long term in the regional(synoptic) climatic conditions of the city and the neighbourhood.

In this chapter, we examine the role of policies and other factors affecting digital technology (DT) integration in mathematics education. In particular, we develop a cross-national analysis of the impact on DT implementation in four countries: two countries in Europe (Italy and Portugal) and two countries in Latin America (Colombia and Mexico). We analyze the role that policies, political changes, reforms, curricula, educational organization and systems, sociocultural aspects, and teachers’ training, knowledge, and beliefs play toward possible DT implementations. We observe that there is a discourse in policies to promote digital technologies’ use, but in practice the availability and integration of such resources in mathematics classrooms is still scarce. We also note that the efforts done during the pandemic did not change this, promoting general ICT use, rather than DT resources that might enhance mathematics teaching and learning.

The adoption of I4.0’s technologies in the work cells accompanied by societal changes calls for new approaches to manage the current production systems. Nevertheless, the current models simulating the behavior of the work cells limit the representation of the operators to the average human, without regard for their individual characteristics, cognitive abilities or psychosocial state. The aim of this paper is to achieve two objectives: firstly, the authors propose a conceptual model for enhancing a human-centered production environment. Secondly, the paper summarizes how the literature characterizes the different dimensions of relations between cognitive and organizational factors. By integrating the collaborative, psychological, social, cognitive, organizational and system performance dimensions, the proposed model focuses on the relationships between these dimensions. Thus, operational models should be closer to the real production environment to improve the design choices of the manufacturing systems.
